First, this method is already in use (e.g.,
http://netaddress.usa.net/) The e-mails that I am getting via this
forwarding service are sponsored by some companies. Also, HotMail adds
"Get your FREE HotMail address" at the end of each letters, sent from
there. It's just easier for a sponsor to pay for sending a message to
10,000 recipients than dealing with hand-crafted work. However, Shawn
proposed exactly that: sending ads to large mailing lists.

.sig's can be made content-specific. It's the writers responsibility
to find an appropriat thread for sponsor's copy.
It depends on the skills of the writer and the content.

We rotate banners to serve more sponsors and to annoy less visitors.
Many people change their sigs depending on the conversation as well.
Something like:

Sarah,
in a gloomy mood

___
Sarah,
listening to Joshua Redman

Believe or not, I've found this Joshua on the Web exactly this way.
Sarah even did not care about the Web. She was listening to her
CD. And I found the site from search engines.

Here is P&G example:

The world is not limited with P&G. As long as inpolite people pollute
newsgroups and our personal mailboxes for free, there should be some
nice people who sponsors posts (and, perhaps, even e-mails).

The last example of content-dependent signature (never used before and
not sure that it will work):
